The story follows Peter Parker, a young boy who discovers his father’s study has been burglarized. His parents gather important documents, leave him with his Aunt May and Uncle Ben, and mysteriously disappear. Years later, Peter, now a teenager, attends Midtown Science High School. Despite his intelligence, he struggles socially and faces bullying. He develops a crush on Gwen Stacy, who shares his feelings.
Peter learns that his father worked with Dr. Curt Connors at Oscorp in the field of cross-species genetics. After sneaking into Oscorp, Peter is bitten by a genetically altered spider, granting him spider-like abilities such as super-strength, heightened senses, agility, and speed.
While exploring his father’s research, Peter visits Connors, who has only one arm. Peter reveals his identity and provides Connors with the missing piece of his father’s work, a “decay rate algorithm,” essential for Connors’ experiments in limb regeneration.
At home, Peter and Uncle Ben argue, leading Peter to leave. While searching for Peter, Ben is shot by a thief whom Peter had previously allowed to escape. Devastated, Peter uses his newfound powers to hunt down criminals resembling the killer. He creates a mask and suit to conceal his identity and constructs mechanical web-shooters from wristwatches.
Peter’s relationship with Gwen deepens, but he discovers her father, George Stacy, is a police captain who opposes the emerging vigilante. Despite this, Peter reveals his identity to Gwen, and they share a kiss.
Meanwhile, Connors successfully tests his serum on lab rats using lizard DNA. However, when his superior demands immediate human trials, Connors refuses, fearing the risks. Desperate, he tests the formula on himself, regrowing his missing arm.
The film explores Peter’s journey as he embraces his abilities, confronts personal loss, and battles threats like the transformed Dr. Connors.
