Ashok (Saif Ali Khan) and Kumar (Riteish Deshmukh) are inseparable friends, with Shanaya (Tamannaah Bhatia) and Mishti (Bipasha Basu) as their respective love interests. Ashok’s father, a billionaire, has been in a coma for six years, leaving his scheming uncle, Kunwar Amarnath Singh, also known as Mamaji (Ram Kapoor), to plot against him. Teaming up with the villainous Dr. Khan (Nawab Shah), Mamaji devises a plan to eliminate Ashok and seize his wealth. During a board meeting, Mamaji drugs Ashok and Kumar, causing them to behave like dogs. They are sent to a mental asylum for treatment under Dr. Shivani (Esha Gupta), who uncovers the truth and vows to release them.
However, chaos ensues when two lookalikes of Ashok and Kumar, who work for a cocaine smuggler named Bijlani (Chunkey Pandey), are mistakenly released instead of the real pair. Shanaya and Mishti take the imposters to Ashok’s mansion, where Mamaji spots the opportunity to exploit the mix-up and claim the family fortune. Meanwhile, the real Ashok and Kumar remain trapped in the asylum. With the help of a ward boy named Cyrus (Darshan Jariwala), they discover a secret ward where they meet Johnny, a lookalike of Mamaji with a peculiar trigger: he attacks anyone who sneezes near him.
As Ashok and Kumar plan their revenge with Johnny’s assistance, they face another hurdle when the asylum warden, Y.M. Raj (Satish Shah), catches them. However, a sneeze from the warden triggers Johnny’s aggression, allowing the trio to escape. On the day of the board meeting, Ashok and Kumar attempt to expose Mamaji, but Dr. Khan triggers a fire alarm, causing the asylum patients to flee.
In a final twist, Ashok and Kumar encounter Balbir, a dance bar owner who is another Mamaji lookalike. Meanwhile, Mamaji arranges for yet another set of Ashok and Kumar doubles through plastic surgery, setting the stage for a chaotic showdown.
This hilarious and chaotic comedy is filled with mistaken identities, revenge plots, and unexpected twists, making Humshakals a memorable Bollywood film.
